Sperm immobilization is a process of sperm membrane damage, which is conducted prior to intracytoplasmic sperm injection (ICSI). This is a necessary process for effective fertilization, as it activates oocyte caused by spermatozoon.
Steps of Sperm Immobilization by Intracytoplasmic Sperm Injection (ICSI)
Use of Polyvinylpyrrolidone (PVP) in Sperm Immobilasation:
Intracytoplasmic sperm injection (ICSI) becomes viscous solution with HAS-7%. This viscous solution slowdown or immobilize sperm and allows easier sperm micromanipulation for intracytoplasmic sperm injection (ICSI).
After making the isci dish we aid seven percent of polyvinylpyrrolidone (PVP), and then add sperm into it. The polyvinylpyrrolidone (PVP) helps to make sperm motility slow. This is very important for sperm immobilization. It is usually done before intracytoplasmic sperm injection (ICSI). It also contain hyaluronate it. Haluranate is a substance found naturally in the reproductive tube.
With the help of injection pipette the tail of the sperm is punctured. This is generally followed by morphological selection of sperm. This sperm selection is based on the Kunger’s criteria.
Sperm head size:
Normal sperm head size should be oval and with well defined acrosome. Acrosome that covers 40-70% of the head is considered normal. Sometimes, the oval head of spermatozoa head with no gross abnormalities are considered abnormal, if they don’t match the exact measurement established by kunger’s strict criteria. If there are any abnormalities recorded in the neck, mid-piece or tail but head is normal it is regarded as abnormal.
Sperm production:
The leftover of cytoplasmic drops are produced in the sperm production, but they are only considered normal when their size is not more than half the sperm head. Take the immobilized sperm into the injection pipette to fertilize the oosyte.
